Overview This patent discloses a process for the fermentative preparation of L-lysine using Coryneform bacteria in which the nucleotide sequence which codes for the enzyme transketolase (tkt gene) is amplified, in particular over-expressed. The process involves, A) amplification of tkt gene coding for transketolase in Coryneform bacterium, B) fermentation of the bacterium in which tkt gene is amplified in a medium, C) concentration of L-Lysine in the medium or in the cells of the Coryneform bacterium and D) isolation of L-Lysine.
